あなた have an 時 and 20 分 to do part 2 after the gap of 80 または 100 または whatever 分 between the two halves. If あなた don't start the 秒 part of the potion within that 時 and 20 mins, the potion screws up. So no, あなた can't leave it for the 次 day.

Usually, if あなた go to the section "cauldrons", あなた should see the percentage how far the Potion has progressed.
