She didn't know who Henry was so I doubt she would know who Emma is. Rumple knows but I believe he is the only one who does. Rumple got Henry for Regina because he was Emma's son, "the daughter of Snow and Charming".

honestly before reading you pick i thought she did but then i really thought about it and no i don't think so. but with a lot of stuff in this show i am not 100% sure

I know she know knows Snow and Charming had a daughter who got away to our world, but I think she has some type of idea.

why else would she have been so desperate to get Emma out of town? why else would she hate her so much?

LET'S NOT FORGET: Regina knows nobody can leave nor COME TO STORYBROOKE!

The fact that Emma could enter the town was a major hint don't you think?! Plus: the clock started working since Emma arrived. Regina should be very naive not to know.
